Description

This course equips learners with the knowledge to assess the evidential value of forensic biology evidence in criminal investigations. It covers the collection, preservation, and analysis of biological samples, and their role in the justice system. By the end, learners will understand how to evaluate the integrity and significance of forensic biology evidence.

Course Outline

Day 1: Introduction to Forensic Biology and Evidence Value

Objectives:
• Define forensic biology and its role in criminal investigations
• Explain the concept of evidence value and its determinants
• Identify the types of biological evidence commonly encountered
• Understand the chain of custody and its importance
• Outline the legal framework governing forensic evidence in South Africa

• Overview of forensic biology
• What is evidence value? Factors affecting value
• Types of biological evidence: blood, semen, saliva, hair, tissue
• Chain of custody: principles and documentation
• Legal context: SAQA, SAPS, and forensic standards

Day 2: Biological Evidence Collection and Preservation

Objectives:
• Describe proper techniques for collecting various biological samples
• Explain preservation methods to maintain DNA integrity
• Recognize contamination risks and mitigation strategies
• Demonstrate documentation of evidence collection
• Understand packaging and labeling requirements

Topics:
• Collection methods for different evidence types
• Preservation: drying, refrigeration, chemical stabilizers
• Contamination control: PPE, sterile tools, and techniques
• Documentation: forms, photographs, and notes
• Packaging: paper bags, envelopes, and containers

Day 3: DNA Structure, Function, and Analysis Principles

Objectives:
• Describe DNA structure and function in the context of forensic analysis
• Explain the principles of DNA profiling (STR analysis)
• Understand the steps in DNA extraction, quantification, and amplification
• Interpret basic DNA profiles and allele frequencies
• Discuss the role of DNA databases (e.g., NFDD)

• DNA structure: double helix, nucleotides, and loci
• Forensic DNA profiling: STRs and multiplex PCR
• DNA extraction methods: organic, silica, and magnetic beads
• Quantification and amplification (PCR)
• Interpretation of profiles: homozygotes, heterozygotes, mixtures
• DNA databases: NDIS, NFDD, and ethical considerations

Day 4: Serology and Body Fluid Identification

Objectives:
• Identify presumptive and confirmatory tests for blood, semen, and saliva
• Explain the principles of serological testing
• Perform basic serological screening techniques
• Interpret test results and understand limitations
• Understand the role of serology in evidence triage

Topics:
• Presumptive tests: Kastle-Meyer, Luminol, Acid Phosphatase, Amylase
• Confirmatory tests: RSID, microscopy, and immunological assays
• Serological screening protocols
• Result interpretation: false positives/negatives
• Evidence triage: prioritizing samples for DNA analysis

Day 5: Trace Biological Evidence and Non-Human DNA

Objectives:
• Describe types of trace biological evidence (e.g., touch DNA, hair)
• Explain collection and analysis of trace evidence
• Understand the value of non-human DNA (animal, plant, microbial)
• Discuss forensic botany and entomology applications
• Evaluate the probative value of trace evidence

Topics:
• Touch DNA: collection via swabs and tape lifts
• Hair analysis: microscopic examination and mitochondrial DNA
• Forensic botany: pollen, plant fragments
• Forensic entomology: insect life cycles and PMI estimation
• Non-human DNA databases and case examples

Day 6: Statistical Interpretation and Population Genetics

Objectives:
• Explain the principles of population genetics relevant to forensic DNA
• Calculate random match probability (RMP) and likelihood ratios (LR)
• Understand the impact of population substructure and databases
• Interpret mixture analysis and complex profiles
• Communicate statistical evidence in court

• Hardy-Weinberg equilibrium and allele frequencies
• Random match probability: product rule and correction factors
• Likelihood ratios: principles and calculation
• Mixture interpretation: allele counting, peak height ratios
• Statistical reporting: verbal scales and courtroom presentation

Day 7: Quality Assurance and Laboratory Standards

Objectives:
• Describe quality assurance (QA) and quality control (QC) in forensic labs
• Explain accreditation standards (ISO 17025, SANAS)
• Understand proficiency testing and validation requirements
• Identify sources of error and contamination
• Discuss ethical responsibilities and expert witness testimony

• QA/QC: controls, audits, and corrective actions
• ISO 17025: key requirements for forensic labs
• Validation of methods: developmental and internal
• Contamination prevention and monitoring
• Ethics: objectivity, confidentiality, and expert testimony

Day 8: Case Studies and Courtroom Testimony

Objectives:
• Analyze real-world case studies involving biological evidence
• Evaluate the impact of evidence value on case outcomes
• Prepare expert testimony and handle cross-examination
• Understand admissibility criteria (e.g., Frye, Daubert, S v Shabalala)
• Develop critical thinking for evidence assessment

• Case study 1: Sexual assault and DNA evidence
• Case study 2: Homicide and trace evidence
• Case study 3: Cold cases and DNA databases
• Courtroom testimony: direct and cross-examination techniques
• Admissibility: legal standards and challenges

Day 9: Integration, Review, and Assessment

• Synthesize knowledge from all days into a coherent framework
• Apply evidence value assessment to complex scenarios
• Demonstrate understanding through a written assessment
• Identify future trends in forensic biology
• Provide course feedback and action plan

Overview

Hands-on practical sessions reinforce theoretical concepts by allowing learners to collect, preserve, and analyze biological evidence in a simulated laboratory and crime scene environment. These sessions develop critical technical skills and ensure proficiency in serological screening and DNA profiling techniques.

Practical Activities
  • Practical 1: Crime Scene Simulation - Evidence Collection — Learners process a mock crime scene, identifying and collecting biological evidence (blood, semen, saliva) using proper techniques and documentation. (9h)
  • Practical 2: Serological Screening and DNA Extraction — Learners perform presumptive and confirmatory serological tests on collected samples, then extract DNA from positive samples using commercial kits. (9h)
  • Practical 3: DNA Profiling and Interpretation — Learners amplify extracted DNA via PCR, run capillary electrophoresis, and analyze resulting profiles to determine matches and calculate statistics. (9h)

Summatives

Each delegate is assessed continuously throughout the course via daily exercises, scored practical assignments, and a final summative test at the end.

Practical Assignments — 30%

Practical assignments are observed and scored against a rubric during the practical sessions. Each delegate's practical mark is averaged into a single 100% score and contributes 30% to the final total.

Daily Exercises — 20%

Every training day ends with a multiple-choice exercise scored out of 100%. The scores from each daily exercise are averaged across the duration of the course to produce a Daily Average mark, which contributes 20% to the final total.

Final Test — 50%

On the last day a final summative test is written. It is a multiple-choice paper with multiple-answer questions: each question may have more than one correct option, and a single wrong selection on a question marks the entire question wrong — no partial credit. The final test is scored out of 100% and contributes 50% to the overall mark.

Awarded to delegates who achieve an overall mark of 50% or higher on the Final Total (Practicals 30% + Daily Average 20% + Final Test 50%).
