Day 2: Chromatographic Theory and Separation Mechanisms

Objectives:
• Describe the theory of chromatographic separation
• Understand retention time, selectivity, and resolution
• Explain the role of stationary and mobile phases
• Calculate key chromatographic parameters

Topics:
• Partitioning and adsorption mechanisms
• Retention factor, selectivity factor, resolution
• Van Deemter equation and band broadening
• Column efficiency: theoretical plates and HETP
• Temperature programming principles

Day 3: GC Instrumentation – Injectors and Columns

Objectives:
• Differentiate between split/splitless, on-column, and PTV injectors
• Select appropriate column types and dimensions
• Understand column stationary phases and their selectivity
• Optimize injection parameters for sample integrity

• Injector types and operation
• Capillary vs. packed columns
• Stationary phases: non-polar, polar, and specialty phases
• Column dimensions: length, ID, film thickness
• Column installation and conditioning

Day 4: Detectors in Gas Chromatography

Objectives:
• Compare common GC detectors (FID, TCD, ECD, MS)
• Select detector based on analyte properties
• Understand detector response and linearity
• Interpret detector signals for quantification

• Flame ionization detector (FID): principle and applications
• Thermal conductivity detector (TCD): universal detection
• Electron capture detector (ECD): halogenated compounds

Day 5: Method Development and Optimization

• Develop a GC method for a given sample matrix
• Optimize temperature program and flow rate
• Select appropriate injection and detection conditions
• Evaluate method performance criteria

• Carrier gas selection and flow optimization
• Split ratio and injection volume adjustment

Day 6: Qualitative and Quantitative Analysis

Objectives:
• Identify compounds using retention times and spectral libraries
• Perform quantitative analysis using calibration curves
• Apply internal standard and standard addition methods
• Calculate concentrations and report results

• Qualitative analysis: retention time matching and Kovats indices
• External standard calibration

Day 7: Sample Preparation Techniques for GC

Objectives:
• Describe common sample preparation methods for GC
• Perform liquid-liquid extraction and solid-phase extraction
• Understand derivatization to increase volatility
• Apply headspace and SPME for volatile analysis

Day 8: Troubleshooting and Maintenance of GC Systems

• Identify common GC problems and their causes
• Perform routine maintenance on injector, column, and detector
• Diagnose issues from chromatogram anomalies
• Implement corrective actions to restore performance

• Common issues: ghost peaks, baseline drift, tailing
• Injector maintenance: septum, liner, syringe
• Column care: cutting, conditioning, storage
• Detector maintenance: cleaning, replacement
• Leak checking and gas purity

Day 9: Data Analysis and Interpretation

Objectives:
• Use chromatography data software for analysis
• Interpret complex chromatograms with multiple peaks
• Apply statistical tools for data quality assessment
• Generate reports meeting regulatory requirements

• Peak deconvolution and resolution enhancement

Day 10: Advanced Applications and Course Review

Overview

Hands-on practical sessions are essential for learners to gain proficiency in GC operation, method development, and troubleshooting. These sessions allow learners to apply theoretical knowledge to real samples, operate instrumentation, and interpret results under supervision.

Practical Activities
  • Practical 1: GC System Familiarization and Injection Techniques — Learners identify GC components, set up a basic method, and perform manual injections of a standard mixture. (8h)
  • Practical 2: Method Development and Optimization — Learners develop and optimize a GC method for a mixture of alcohols, adjusting temperature program and flow rate to achieve resolution. (8h)
  • Practical 3: Quantitative Analysis of a Spiked Sample — Learners prepare calibration standards, analyze a spiked water sample, and quantify target compounds using external standard calibration. (8h)
  • Practical 4: Troubleshooting and Maintenance — Learners diagnose common GC issues (e.g., tailing peaks, baseline drift) and perform routine maintenance such as replacing a septum and liner. (8h)

Summatives

Each delegate is assessed continuously throughout the course via daily exercises, scored practical assignments, and a final summative test at the end.

Practical Assignments — 30%

Practical assignments are observed and scored against a rubric during the practical sessions. Each delegate's practical mark is averaged into a single 100% score and contributes 30% to the final total.

Daily Exercises — 20%

Every training day ends with a multiple-choice exercise scored out of 100%. The scores from each daily exercise are averaged across the duration of the course to produce a Daily Average mark, which contributes 20% to the final total.

Final Test — 50%

On the last day a final summative test is written. It is a multiple-choice paper with multiple-answer questions: each question may have more than one correct option, and a single wrong selection on a question marks the entire question wrong — no partial credit. The final test is scored out of 100% and contributes 50% to the overall mark.
